چکیده

The pathophysiology of multiple disease states, including sepsis,1,2 hypertension,1 atherosclerosis,3–5 and ischemiareperfusion injury,6 is characterized by loss of the endothelial barrier function leading to the accumulation of plasma-borne proteins, tissue edema, and cell death. Importantly therapeutic strategies that inhibit loss of endothelial barrier function have been associated with strategies that improve outcomes.7 There is growing understanding of the role of adherens in maintaining the integrity of the endothelial barrier function8 and the molecular pathways that regulate adheren expression.9

Bench-to-bedside review: The role of activated protein C in maintaining endothelial tight junction function and its relationship to organ injury

Activated protein C (APC) has emerged as a novel therapeutic agent for use in selected patients with severe sepsis, even though the mechanism of its benefit is not well established. APC has anticoagulant, anti-inflammatory, antiapoptotic, and profibrinolytic properties, but it is not clear through which of these mechanisms APC exerts its benefit in severe sepsis. Focus has recently turned to th...

To be or not to be protease activated receptor-1 in activated protein C-initiated endothelial barrier protection?

The review by Looney and Matthay summarized recent progress in basic and preclinical research of activated protein C (APC), a novel therapeutic agent for the treatment of severe sepsis [1]. APC is traditionally an anticoagulant and profibrinolytic agent, and has been appreciated to possess anti-inflammatory and anti-apoptotic properties, yet the exact mechanisms by which APC executes its clinic...

Unraveling the mechanisms involved in endothelial barrier protective effects of angiopoietin-1 variant MAT.Ang-1

eri and colleagues [1], demonstrating that angiopoietin (Ang)-1 variant MAT.Ang-1 improved endotoxemiainduced microvascular dysfunction and microvascular hyperpermeability. Th e authors suggested that MAT. Ang-1-induced recovery of microcirculatory tissue perfusion during sepsis is due to preservation of endothelial barrier integrity. To further elucidate the mechanism, they investigated the po...

Sepsis-induced elevation in plasma serotonin facilitates endothelial hyperpermeability

Hyperpermeability of the endothelial barrier and resulting microvascular leakage are a hallmark of sepsis. Our studies describe the mechanism by which serotonin (5-HT) regulates the microvascular permeability during sepsis. The plasma 5-HT levels are significantly elevated in mice made septic by cecal ligation and puncture (CLP). 5-HT-induced permeability of endothelial cells was associated wit...

LIM kinase 1 promotes endothelial barrier disruption and neutrophil infiltration in mouse lungs.

RATIONALE Disruption of endothelial barrier function and neutrophil-mediated injury are two major mechanisms underlying the pathophysiology of sepsis-induced acute lung injury (ALI). Recently we reported that endotoxin induced activation of RhoA in mice lungs that led to the disruption of endothelial barrier and lung edema formation; however, the molecular mechanism of this phenomenon remained ...
